The unknown substance contained glucose, starch, and proteins. We know that the substance contained glucose because when we added the Benedict Solution, the substance turned from its initial color blue to it’s final color orange meaning the food sample reacted to the solution. We also know that starch was present in the substance because when we used the Lugol 's solution the substance reacted and turned from its initial color yellow to its final color blue/black. Lastly we know that the substance contains Proteins because of its reaction with Biuret solution. When the substance reacted with the solution it turned from its initial color yellow/brown to its final color lilac/violet. The experiment went by easily flowing nicely, although one or two things went wrong, none had any effect on the experiment. It is very important to know what foods are composed of because, knowing what is inside our food is essential for our health. We need to be aware of what things we are consuming and what we should be consuming for our bodies to function properly. An application for testing what foods are composed of is for health reasons. It was also to see how you can identify which biomolecules are present in a substance or a food. I think we achieved the purpose of this experiment and fulfilled what it intended us to do. As we come to the conclusion, I have learned that my hypothesis is indeed correct and there was glucose,starch, and proteins present in the unknown substance. My lab group personally had no idea what the substance was before or after the experiment took place, because glucose, starch, and proteins are present in many other things so learning that these molecules were present didn’t really help us find out what the substance
Fermentation test is used to determine if unknown #398 uses any oxygen to ferment carbohydrates and acids. Oxidation tests were used to determine if unknown #398 metabolizes carbohydrates and acids by cellular respiration. Both tests are observed by inoculation of unknown #398 into 3 sugar broths: lactose, glucose, and mannitol and 1 citrate (Citric acid) slant. Fifth test, Hydrolytic and Degradative reactions is used to determine if unknown #398 contains enzyme, amylase that hydrolyzes starch after streaking on a starch plate. Next test, inoculation of a urea broth and is used to determine if unknown #398 contains urease that hydrolyzes urea.
First, it was hypothesized that test tube "A", the control, would not show any red concentration, test tube "B" which contains supernatant II would show the most red concentration and test tube "C" which contains sediment II would only show a little red concentration. The second hypothesis states that the raw corn kernels would have mitochondrial activity while the boiled corn kernels would not. The last hypothesis interprets that the "gunk" and sediment I will both contain starch granules. It was only expected to find mitochondrial activity in Supernatant II. Unfortunately, after performing this experiment, we were not able to support this hypothesis and come up with a conclusion.

Starch solution is then placed into the test tube at a quantity of 5 mL. 5 drops of Lugol’s Iodine solution is added to the test tube. If the color changes, then it is known that starches are present in the solution. Proteins are next tested. In order to do this, 5 mL of gelatin solution is added to the test tube. 10 drops of Biuret’s reagent are added to test for protein.
These color changes indicate a chemical change, which show that a reaction had occurred. In the first step when o-vanillin and p-toludine, imine was formed. The color change from green to orange suggests that imine appears as orange colored. In the second step, the addition of sodium borohydride reduced the imine into another derivative, which was yellowish lime color. The solution turned clear when acids and anhydrides was added, which indicated the precipitate were dissolved.
The investigation was carried out to identify the presence or absence of biological molecules in serum 2216. If the concentration in each test tube of the dilutions carried out will be more concentrated then the concentration of the test tube before it, then the color will be at an equal concentration with the other dilutions performed. The hypothesis was wrong because of the difference in concentrations due to the different measurements within the dilutions done. The test for starch was to add a drop of iodine solution to the pipette in the spotting tile. A reducing sugar solutions is add inside a test tube with 3 drops to then add 3 drops of benedicts and plane in a water bath.

- A hydrate is a salt that contains water as a part of its crystal structure. The hydrate used in this lab was Copper (ll) Sulfate Pentahydrate. To heat the hydrate in this lab a crucible is needed. A crucible is a heat resistant container used to heat things to high temperatures. In this lab a mole was used to determine the measurements of all substances.
A larger sample size could have enabled averages to be resulted, and the experiment could have had several trials until concordant results are attained to increase the reliability. To enhance the accuracy of the results, a machine such as a cheese wire to cut straight along the edge could have been employed that would more accurately cut the cubes and hence reducing human error. This would be efficient as there is a large chance of human error when cutting the cubes by hand as the agar is slippery. Cubes with different sizes could be observed to improve the validity of the experiment. The weight of the bottom of the cubes pushing against the beaker likely caused an error to occur.
The iodine test determines the presence of starch in biological materials. It is predicted that, if starch is not present, the solution with iodine remains yellow. However, if starch is present the solution with iodine becomes a blue-black colour. Plants have starch as the storage polysaccharide (glucose units held together by glycosidic bonds) while animals have the equivalent of glycogen. In this experiment, the dark blue colour is visible because of the helical amylose and amylopectin reacting with iodine (Travers et al., 2002).

The experiment is conducted by laboratory method (Biuret Test) and using spectrophotometer to analyze the absorbance of reactants at 540 nm, hence determining the concentration of the proteins in a given sample. The purpose of stopped enzyme assay to study B-galactosidase is to determine the effect of temperature and concentrations of substrate on enzyme activity.
Analysis Conclusion Based of the data collected from the experiments, cornstarch was identified as the unknown powder in the black bag. Cornstarch shares similar chemical properties compared to the unknown powder. They both have a pH of 7, contain starch, but not carbonate, and react with the flame to create a temporary bright orange. Even so, powdered sugar also has the same chemical properties. However, when the powders were viewed in the Scanning Electron Microscope, the powdered sugar grains looked nothing like that of the unknown sample.
