The Unknown Identification Lab was an experiment that provided the opportunity to apply all the tests that were learned in the semester of lab, to identify the two bacterias that remain unknown. Gram- staining and two other tests will be used to identify the unknowns. This experiment is crucial to the understanding of each test, and can benefit in the ability to identify the characteristics of specific bacteria. Having a clearer understanding of the bacteria can further the research of bacteria for medicine, such as antibiotics. The understanding can also help the development of research in the environment. For example, Clostridium botulinum can cause an issue in can foods. By knowing the previous information, one can avoid certain cans and prevent sickness. Each bacteria has specific characteristics that is unique to the certain strain, which makes identification possible. This Unknown Identification Lab was performed in a series of three days. The lab contained the following tests: Gram-staining, Blood Agar Plate, Mannitol Salt Agar, Levine Eosin Methylene Blue Plate, and Hektoen Enteric Agar Plate. “ Christian Gram was the one who tried to develop the procedure of Gram staining by attempting to create a way to differentiate stains of schizomycetes from the other tissue cells”(Bartholomew).The Gram-staining was used to determine the morphology of the bacteria by using the cell wall structure. When the bacteria is viewed under a microscope, morphology was seen. Bacteria can have one of the three basic morphologies. The bacteria was cocci, rod, or spirilla morphology. If the result of the bacteria is purple, the bacteria has a thick peptidoglycan layer. This also means the bacteria is gram-positive. Methods Unknown microbial #398 went through several of tests in order to identify its characteristics when isolated from a urine sample of Doris, a 64- year old patient with a kidney infection. To identify unknown #398, must prepare a working and a reserve stock by the inoculation from a broth culture and by quadrant streaking method on a PEM and EMP plates. The following test procedures were incubated at 37°C for 48 hours for observation and identification for unknown #398. The identification of unknown #398 followed test procedures from Brown1.

The purpose of this lab report is to employ a myriad of skills, tools and, methods learned throughout this semester to perform the appropriate tests for the identification of the assigned unknown bacteria. Aseptic technique is an important tool for microbiologists. It is imperative that aseptic technique is maintained throughout the length of any test to avoid any cross-contamination that may lead to inaccurate results.

Staphylococcus epidermidis is the organism that was identified based on the tests that I had conducted. The tests that I used to identify this organism were the coagulase test and the catalase test. My bacterium was beta hemolytic as well. First, a gram stain had to be done to determine whether the organism was a gram positive organism or a gram negative organism. This determined which set of tests that had to be done.
These could range from education to becoming more Earth friendly. We can reform education in many ways. We can first start off children with more difficult curriculum in lower grades to get them more prepared for harder and more important subjects. Another way to reform in the classroom is to provide better teachers who will ensure that every student will learn what they are supposed to. On April 6, 2016 at approximately 11:45am, a local police station got a call about a hostage situation at a local pharmacy. When police and medical examiners got to each crime scene, they learned that all of the hostages were given drugs and had overdosed on them. Some of the pills, in powder form, were found near the victims. One of the victims was stable enough to tell the investigators that the power on the floor were the drugs they were forced to take. The medical examiner found out each hostage was given either unknown A or unknown B.
The investigation was carried out to identify the presence or absence of biological molecules in serum 2216. If the concentration in each test tube of the dilutions carried out will be more concentrated then the concentration of the test tube before it, then the color will be at an equal concentration with the other dilutions performed. The hypothesis was wrong because of the difference in concentrations due to the different measurements within the dilutions done. The test for starch was to add a drop of iodine solution to the pipette in the spotting tile. Tests that are used during this experiment were a flame test, ion test, pH test, and conductivity test. The results drawn from these tests confirmed the identity of the Unknown White Compound to be sodium acetate (NaC2H3O2) because there were no presence of ions and sodium has a strong persistent orange color. The compound then will be synthesized with the compounds Na2CO3 and HC2H3O2 to find percent yield.
Aseptic technique was initiated at the beginning of this experiment by cleaning the work surface with disinfected wipes. Personal protectives equipment was also worn. The material utilized in this experiment was: S. epidermidis culture broth, sterile cotton swab, streak plate, forceps in 70% alcohol, a lit tea light, and the three antibiotic disks (novobiocin, gentamicin, penicillin). The first step, I divided a plate into three quadrants and labelled them with the different antibiotic names. Using the lit tea light, like a bursen burner, I flamed the mouth of the S. epidermidis culture.
